1.試劑盒應按標簽說明書儲存，使用前恢復到室溫30分鐘后方可使用。稀稀過后的標準品應丟棄，不可保存。

2.實驗中不用的板條應立即放回包裝袋中，密封保存，以免變質。

3.不用的其它試劑應包裝好或蓋好。不同批號的試劑不要混用.請在試劑盒保質前使用。

4.使用一次性的吸頭以免交叉污染，吸取終止液和底物A、B液時，避免使用帶金屬部分的加樣器。

5.使用干凈的塑料容器配置洗滌液。使用前充分混勻試劑盒里的各種成份及樣品。

6.洗滌酶標板時應充分拍干，不要將吸水紙直接放入酶標反應孔中吸水。

7.底物A應揮發，避免長時間打開蓋子。底物B對光敏感，避免長時間暴露于光下。避免用手接觸，有毒。實驗完成后應立即讀取OD值。

8.加入試劑的順序應一致，以保證所有反應板孔溫育的時間一樣。

9.按照說明書中標明的時間、加液的量及順序進行溫育操作。

樣本保存條件：

1.2-8℃冰箱*多放2周

2.-20到-50℃的冰箱可以放3-6個月

3.-50℃以下的冰箱，可以放一年

樣本收集

1.血清樣本用干燥管（紅色）或者促凝管（黃色）收集

2.血漿用抗凝管，一般有EDTA抗凝（紫色）、枸櫞酸鈉抗凝（黑色或者藍色）

3.動物組織勻漿（細胞）：

              1g組織加入9ml PBS（濃度為0.1M，pH為7.4的）測分泌型指標

              1g組織加入1ml組織裂解液加入8ml PBS 測細胞內

糞便樣本：1g樣本+9ml PBS（濃度為0.01M，pH為7.4的）

沒有PBS的話也可以用一下生理鹽水，還是用PBS

樣本離心轉速及時間

1.試管采集的樣本：3000轉，離心五分鐘取上清

2.離心管或者EP管采集的樣本：5000－－8000轉3分鐘取上清

3..樣本保存－－如果樣品不立即使用，應將其分成小部分-70 ℃保存，避免反復冷凍。盡可能的不要使用溶血或高血脂血。如果血清中大量顆粒，檢測前先離心或過濾。不要在37℃或更高的溫度加熱解凍。應在室溫下解凍并確保樣品均勻地充分解凍。

 定量測定

　　ELSIA操作步驟復雜，影響反應因素較多，特別是固相載體的包被難達到各個體之間的一致，因此在定量測定中，每批測試均須用一系列不同濃度的參考標準品在相同的條件下制作標準曲線。測定大分子量物質的夾心法ELISA，標準曲線的范圍一般較寬，曲線點的吸光度可接近2.0，繪制時常用半對數紙，以檢測物的濃度為橫坐標，以吸光度為縱坐標，將各濃度的值逐點連接，所得曲線一般呈S形，其頭、尾部曲線趨于平坦，中央較呈直線的部分是*理想的檢測區域。

　　測定小分子量物質常用競爭法，其標準曲線中吸光度與受檢物質的濃度呈負相關。標準曲線的形狀因試劑盒所用模式的差別而略有不同。ELISA測定的標準曲線注意圖中橫坐標為對數關系，這更有利于測定系統的表達。
